Lane was born in New York City on 13 November 1933 to a Jewish father (Jacob) and a Catholic mother (Dolly), who eventually converted to Judaism. Dolly was a housewife, and Jacob “Jack” Isaacson was a sergeant in the New York City Police Department (NYPD). Morton (Don) grew up in The Bronx and went to DeWitt Clinton High School, where he was classmates with Judd Hirsch and Garry Marshall. Because of his mother’s dominant personality, Isaacson had determined he wanted to be in the show industry by his mid-teens. He even hung a star on the door of his bedroom. He began to perform at gatherings he attended with his parents, generally by combining singing and humor. DeWitt Clinton High School also had him play varsity football and basketball. He then earned a football and basketball scholarship at Kalamazoo College in Kalamazoo, Michigan, where he stayed for three years.

- After graduating from college, Isaacson established “Donny and the Duke,” a duo with his closest buddy Manny Glasser. For a few months, the pair worked in New York nightclubs until Isaacson was inducted into the US Army at the age of 21.
- The duo went on to win the All-Army entertainment tournament, landing them a spot on The Ed Sullivan Show in 1955. Isaacson won the All-Army entertainment contest as a solo act a year later. Following his necessary army duty, he traveled for two years, entertaining soldiers all throughout the United States.
- Lane also co-hosted a morning radio show with Bert Newton on Melbourne’s 3UZ radio station during his stint on The Don Lane Show. Newton shares a tale about how he and the producer pretended to be on the radio and revealed the listener’s Lane’s home address.
